摘要 本研究延續92、93兩年度研發之技術,將幾丁聚醣(chitosan)固定於接枝聚丙烯酸之聚丁基烷酯(PHB)薄膜表面,並探討各反應條件對固定量之影響。固定反應之進行,乃利用1-ethyl-3-(3-diethylaminopropyl) carbodiimide hydrochloride (EDAC)為媒介,使幾丁聚醣之胺基與丙烯酸反應形成醯胺鍵結,而將其固定於薄膜上;醯胺鍵結之存在則利用FTIR-ATR光譜分析加以確認,固定量受反應條件如EDAC濃度、反應pH值、反應時間、幾丁聚醣濃度、聚丙烯酸接枝量等影響,於最佳條件下,每平方公分之自由胺基數目可達0.26mole,高出文獻所報導數據達200倍之多;而接觸角量測中可發現,薄膜表面經固定幾丁聚醣後,接觸角大幅下降而呈現親水現象。 In this research, the techniques established in previous two years (2003, and 2004) were further applied to immobilize chitosan on PHB membranes that have been grafted with poly(acrylic acid). Immobilization of chitosan on the membranes was achieved by forming amide bonds between amino groups of chitosan with grafted poly(acrylic acid), using 1-ethyl-3-(3-diethylaminopropyl) carbodiimide hydrochloride (EDAC) as an intermediate medium. Existence of amide groups on the formed membrane was confirmed by using FTIR-ATR analysis. The immobilization yields were found to be dependent on reaction conditions, such as concentration of EDAC, pH, reaction time, concentration of chitosan, PAA density, etc. Under an optimal consition, for which the highest attainable yield was obtained, the surface density of free amine reached as high as 0.26 mole/cm2, which value was ca. 200 times that of the data reported in the literature. Firthermore, water contact angle measurements indicated a considerble decrease of the contact angle, approaching an hydrophilic behavior, after the membrnae was immobilized with chitosan.
