摘要 本研究擬分三年時間, 以Serratia ureilytica TKU013所生產生理活性成分為主,及 實驗室已篩選到的其他Serratia屬細菌,分別比較其發酵上清液之生理活性及結構鑑 定。所擬探討的碳氮源包括蝦殼、蟹殼、烏賊軟骨以及其他各種幾丁質及幾丁聚醣產 品(不同去乙醯度、分子量)。 『第一年』擬針對Serratia spp.所生產抗氧化劑進行其最適生產條件之探討,並 分析抗氧化活性之機制及其性質之研究,再比較Serratia ureilytica TKU013與其他 Serratia spp.菌株之發酵上清液其抗氧化活性之差異。 『第二年』擬探討抗氧化物成分之萃取、分離及化學結構之鑑定,並將純化後之 成份分別進行抗氧化及抗腫瘤評估。 『第三年』擬探討有效成份之抗腫瘤機轉分析及有效成份大量生產方法之評估。 此類研究成果得以應用於諸如保健食品及化妝品之類的生物技術或生物醫學材料方面 之用途。 This is a three years project. Several bacterial strains, Serratia ureilytica TKU013 and other Serratia spp. were used for the production of novel physiologically bioactive compounds. The chitinous materials used as the sole carbon/nitrogen source for production of physiologically bioactive compounds were shrimp shell, crab shell, squid pen, chitin, and chitosan. In the first year, the production conditions of antioxidants by Serratia spp. will be studied. The various antioxidant activities of these culture supernatants will be analyzed and compared. In the second year, purification and structural identification of these antioxidants will be studied. The antitumor activities of the pure compounds will also be analyzed. In the third year, the mechanism of the physiologically bioactive compounds to tumor cells will be studied in detail. With these methods, we will show that shrimp and crab shell wastes can be utilized and reveal its hidden potential in the production of functional foods, cosmetics and biomedical materials.
